چکیده

Abstract Preimplantation genetic testing of embryos offers the opportunity to avoid transmission hereditary disorders offspring by selecting out affected a single gene mutation or mutations (PGT-M), structural rearrangements chromosomes (PGT-SR), aneuploidy (PGT-A) as well polygenic risk (PGT-P). Over last two decades, laboratory techniques have seen tremendous developments including extended embryo culture blastocyst stage, trophectoderm biopsy and vitrification. The latter technique is highly efficient allows multiple freezing warming cycles when performed in proficient laboratories. At same time whole genome analysis provide analyse embryonic DNA great detail for indications simultaneously. PGT process sensitive both biological technical issues which sometimes lead failed result. Failure diagnosis reported occur between 2-12% depending on methodology used. main causes inconclusive are amplification failure non-concurrent results. Independent variables that influence results day biopsy, number cells taken at quality IVF laboratory. In order seize every reaching optimize reproductive outcome patient, rebiopsy retesting (double double cryopreservation) can be performed. situations where need biopsied reasons newly found clinical PGT-A, after an initial cryopreservation, cryopreservation may with potential effect subsequent outcome. Isolation precautions are intended to minimize pathogen transmission and reduce hospital-acquired infections. More recently, the effectiveness of isolation precautions has been questioned because of increasing evidence of risks. These putative downsides are divided into a quantifiable monetary cost (i.e., a literal cost to the system) and clinically important but less easily quantifiable costs ...

Tolerance--is it worth it?

We are entering an exciting time in the study of immunologic tolerance. Several cellular and molecular strategies have been developed that show promise in nonhuman transplant models and these approaches are just now appearing in clinical trials. Tolerance strategies that prevent immune rejection and obviate the need for immunosuppressive medications (with inherent risk of cancer, infection, and...
